响应面法优化藏药蕨麻总生物碱提取工艺及其抗氧化研究
Optimization of Extraction Process and Antioxidant Studies of Total Alkaloids from Tibetan Medicine Dryopteris by Response Surface

摘要(Abstract):

为优化蕨麻总生物碱的提取工艺并对其抗氧化活性进行测定,以蕨麻总生物碱提取量为指标,将影响生物碱提取的因素(提取时间、提取温度、溶剂浓度、料液比等)采用单因素实验和Box-Behnken响应面法进行优化。结果最优提取工艺为时间74.93 min、温度75℃、乙醇体积分数90%、料液比1∶30。在此条件下测得蕨麻总生物碱含量为2.324 mg·g~(-1),平均加样回收率为97.967%,RSD=1.35%。DPPPH和ABTS自由基反应结果表明,0.02~0.10 g/L蕨麻总生物碱对DPPPH有清除作用,对ABTS自由基有明显的清除效果。
In this work, we report a study to optimize the extraction process of total alkaloids from Potentilla anserina and then to determine their antioxidant activity. By taking the extracted amount of total alkaloids from Potentilla anserina L. as the descriptor, to optimize the extraction process, we determined and analyzed the impacting factors such as the extraction time, extraction temperature, solvent concentration and solid-to-liquid ratio. The extraction process was finalized by adopting the single factor experiment and the Box-Behnken response surface. According to the software analysis results, the optimum extraction process was determined to the following: extraction time of 74.39 min, at temperature of 75 ℃, with the alcohol volume fraction of 90% and solid-to-liquid ratio of 1∶30. With this condition, the measured content of total alkaloids in Potentilla anserina L. was 2.324 mg·g~(-1), the average spotting recovery rate of 97.967%, and the RSD of 1.35%. Our results of antioxidant studies with DPPPH and ABTS free radicals showed that 0.02~0.10 g·L~(-1) Potentilla anserina L. alkaloid exhibited significant scavenging effects on DPPPH and ABTS free radicals.
